Cryopreservation of Plasma

Sucrose solution for the cryopreservation of plasma (Ref: Rumsey et al. Journal of lipid res. 1994)

For a 50ml solution:

  • In an LPS free clean vessel add
    • LPS free Sucrose 0.25g
    • LPS free NaCl 0.48g
    • LPS free EDTA 4.7mg
  • Add 40ml pyrogen-free water to dissolve
  • Adjust pH to 7.4
  • Bring to 50ml, sterile-filter, aliquot in 1 ml tubes. Keep sterile at 4°C. Do not reuse. LPS interferes with your experiments.
  • Add 1:4 (v:v) one volume sucrose solution : to four volume plasma, mix very very well. Wrap Parafilm at the top, and keep the sample frozen.

This solution contains:

  • 50% Sucrose
  • 150mM NaCl
  • 0.24mM EDTA
