Sucrose solution for the cryopreservation of plasma (Ref: Rumsey et al. Journal of lipid res. 1994)
For a 50ml solution:
- In an LPS free clean vessel add
- LPS free Sucrose 0.25g
- LPS free NaCl 0.48g
- LPS free EDTA 4.7mg
- Add 40ml pyrogen-free water to dissolve
- Adjust pH to 7.4
- Bring to 50ml, sterile-filter, aliquot in 1 ml tubes. Keep sterile at 4°C. Do not reuse. LPS interferes with your experiments.
- Add 1:4 (v:v) one volume sucrose solution : to four volume plasma, mix very very well. Wrap Parafilm at the top, and keep the sample frozen.
This solution contains:
- 50% Sucrose
- 150mM NaCl
- 0.24mM EDTA
